There are plenty to consider. Ajax for one, which I prefer to jquery .....

I would recommend visiting a site called webdeveloper.com ... State the rough project in mind .... some seasoned web developers there .....

Brush up on your "object-oriented" Javascript if you are going to be using js frameworks.

Not to be pedantic but Ajax is a group of methods wheras jQuery is a javascript library so they cant be compared, an implementation of Ajax may or may not include jQuery.

It won't be public facing web stuff, more in house stuff with a web front end.
While you're waiting, read the free novel we sent you. It's a Spanish story about a guy named 'Manual.'

Hmm. I've had a tinker with EC2 (was thinking of using it to host instances of my pet project / plan B) and I use google docs and stuff quite a bit. Anything else I should be worrying about?

They may be referring the to MS ASP.NET Ajax Library. That is one I know pretty well however my preference lies with jQuery. Additionally, it looks like jQuery is more beneficial to your CV.

There is a relatively new JS library called "Knockout JS", not sure what the up take will be on that one but I'm tempted to use it on the next project if it's relevant.
